Matrix-M is a saponin-based vaccine adjuvant composed of purified saponins from *Quillaja saponaria*, cholesterol, and phospholipids, forming 40 nm cage-like nanoparticles. It enhances both humoral and cellular immunity by stimulating the innate immune system and improving antigen presentation, leading to stronger and broader antibody responses and a balanced Th1/Th2 T cell profile. It has a favorable safety profile in clinical trials and allows for dose-sparing of antigens in vaccines.
Stimulates the innate immune system via recruitment and activation of immune cells (neutrophils, DCs, macrophages, T-cells, B-cells, and NK-cells) at the injection site and draining lymph nodes, leading to enhanced antigen presentation and adaptive immune responses.
