Matrix metalloproteinase-11 (MMP11), also called Stromelysin-3, is a zinc-dependent endopeptidase and member of the matrix metalloproteinase family, involved mainly in the remodeling of the extracellular matrix. Uniquely among MMPs, MMP11 is activated intracellularly and does not efficiently degrade major structural ECM proteins but instead targets specific proteins, such as protease inhibitors and insulin-like growth factor-binding protein-1. MMP11 is highly expressed in various cancers, contributing to tumor cell proliferation, invasion, metastasis, and resistance to apoptosis, making it a promising therapeutic target and biomarker in oncology. MMP11’s activity shapes the tumor microenvironment and correlates with poor prognosis in multiple solid tumors. Antibody-based therapies and cancer vaccines targeting MMP11 are in preclinical or early clinical investigation[1][2][3][4][5][6].
