Overview

Matrix-remodeling-associated protein 5 (MXRA5) is a large secreted glycoprotein (312 kDa) encoded by the MXRA5 gene in humans, featuring 7 leucine-rich repeats and 12 immunoglobulin-like C2-type domains related to perlecan[1][2][4][5]. It is primarily involved in extracellular matrix remodeling and cell adhesion in various tissues, mediating processes such as cell proliferation, cell cycle regulation, and epithelial-to-mesenchymal transition (EMT)[1][2][3]. MXRA5 is upregulated in disease states such as cancer (notably pancreatic, colorectal, and non-small cell lung cancer), BPH, osteoarthritis, and chronic cardiac and renal disorders[2][3][7]. It exerts anti-inflammatory and anti-fibrotic effects in the kidney and modulates signaling pathways including MAPK (ERK1/2, p38) and PI3K-Akt-mTOR[1][2][5].
