The Matrix remodeling-associated protein 5 (MXRA5) peptide–HLA class I complex is a tumor-associated antigen (TAA) target utilized in cancer immunotherapy. MXRA5, also known as adicardin, is a large glycoprotein involved in extracellular matrix remodeling and is frequently overexpressed in various malignancies, including glioblastoma, colorectal cancer, and non-small cell lung cancer [1][2]. In these cancer cells, MXRA5 is proteolytically processed into short peptides that are subsequently loaded onto Human Leukocyte Antigen (HLA) class I molecules, such as HLA-A*02:01, and presented on the cell surface [3]. This specific peptide-HLA complex serves as a ligand for the T-cell receptors (TCRs) of CD8+ cytotoxic T lymphocytes, which can be activated to recognize and kill the tumor cells [4]. Therapeutic interventions targeting this complex primarily include multi-peptide vaccines like IMA950 and IMA910, which aim to prime the patient's immune system to mount a specific attack against cells displaying the MXRA5 antigen [5]. The clinical utility of this target depends on the high differential expression of MXRA5 in tumors compared to healthy tissues and the presence of the appropriate HLA allele in the patient [2][4].
