Overview

The Matrix-remodeling-associated protein 5 (MXRA5) peptide-HLA complex is a tumor-associated antigen (TAA) target utilized in advanced cancer immunotherapies. MXRA5, also known as Adicartin, is a secreted protein involved in the remodeling of the extracellular matrix and is significantly overexpressed in several cancers, such as non-small cell lung cancer (NSCLC), colorectal cancer, and glioblastoma (UniProt Q9NR99). The target specifically refers to a processed peptide derived from MXRA5, such as the MXRA5-001 epitope, presented on the cell surface by Human Leukocyte Antigen (HLA) molecules, most commonly HLA-A*02 (PMID: 22016301). This complex is recognized by the T-cell receptors (TCRs) of cytotoxic T-lymphocytes, which then mediate the lysis of the presenting tumor cells. Therapeutic strategies targeting this complex include multi-peptide vaccines like IMA950 and adoptive cell therapies like IMA101, which aim to stimulate or provide a robust immune response against MXRA5-positive tumors (Immatics N.V.). Because MXRA5 expression is relatively restricted in healthy adult tissues, it offers a favorable therapeutic window, although monitoring for off-target effects remains a standard safety consideration in clinical trials. The complex serves as a critical bridge between the intracellular proteome of the cancer cell and the adaptive immune system, allowing for highly specific tumor targeting.
