Overview

Matrix remodeling-associated protein 8 (MXRA8) is a type I transmembrane protein involved in cartilage formation, angiogenesis regulation, and blood-brain barrier maintenance. It also acts as an entry receptor for arthritogenic alphaviruses. In cancer, high MXRA8 expression is associated with poor prognosis in glioma and identifies a subset of cancer-associated fibroblasts (CAFs) in pancreatic ductal adenocarcinoma. Targeting MXRA8 has shown promise in inhibiting tumor cell proliferation and inducing ferroptosis.

Mechanism of action

Inhibition of MXRA8 leads to increased NCOA4 expression and decreased FTH1 levels, promoting ferroptosis. Antibody-based therapies targeting MXRA8.
