MCC regulator of Wnt signaling pathway, abbreviated as MCC, is a nuclear and membrane-associated phosphoprotein that acts as a tumor suppressor, particularly in colorectal cancer, by negatively regulating cell cycle progression and inhibiting the Wnt/β-catenin signaling pathway[1][3][5]. MCC is critical for the maintenance of cytoskeletal integrity and cell survival in podocytes, where its loss leads to structural injury, apoptosis, and downregulation of essential proteins[1]. While primarily functioning as a tumor suppressor, MCC may act as an oncogene in certain B-cell malignancies. In the kidney, it is essential for maintaining the specialized structure of podocytes, and its deficiency is implicated in diseases such as diabetic nephropathy and focal segmental glomerulosclerosis[1]. MCC’s roles are cell type dependent, and while it acts via Wnt/β-catenin inhibition in some contexts, its podocyte functions also relate to cell structure. MCC is not currently known to be directly targeted by any approved drugs, but modulation of the Wnt/β-catenin pathway is a therapeutic strategy in oncology and kidney diseases[1][3][5].
