MDC1 antisense RNA 1 (MDC1-AS1) is a long noncoding antisense RNA transcribed from the opposite strand of the MDC1 gene, which encodes the mediator of DNA damage checkpoint protein 1[3][2]. MDC1-AS1 has been identified as downregulated in various human cancers, including bladder cancer, gastric cancer, and glioma[3][2][5]. The RNA positively regulates its sense coding gene MDC1, known for its role as a tumor suppressor and crucial mediator of the DNA damage response[2][1][3]. Overexpression of MDC1-AS1 leads to increased MDC1 mRNA and protein levels, resulting in suppressed proliferation, migration, and invasion of cancer cells[3][1]. The tumor-suppressive effect of MDC1-AS1 appears to be mediated via upregulation of MDC1; when MDC1 is knocked down, the inhibitory effect of MDC1-AS1 on tumor cell behavior is lost[3][1]. Although MDC1-AS1 is implicated in cancer biology, it is not a receptor, enzyme, or other traditional therapeutic target molecule, but is rather an antisense lncRNA that may regulate the expression of a tumor suppressor gene[3][2][1][5].
