Overview

Mdm1 nuclear protein (MDM1) is a microtubule-binding nuclear protein that localizes primarily to the centrioles of dividing cells and differentiating multiciliated cells[1][2][3][4][5]. It acts as a negative regulator of centriole duplication, likely through direct interaction with and stabilization of microtubules. MDM1's repeat motifs contribute to its microtubule-binding ability. Mutations in MDM1, mainly identified in mouse models, lead to age-related retinal degeneration and are associated with various human ocular diseases, emphasizing the protein's key role in cellular homeostasis and retinal integrity. There is no evidence to support MDM1 being a classical drug target, receptor, enzyme, transporter, or transcription factor; instead, it is best classified as a “microtubule-binding protein” with established roles in centriole biology and retinal pathogenesis, but not as a current therapeutic target[1][2][3][4][5].
