MDM2 is a proto-oncogene encoding a nuclear-localized E3 ubiquitin ligase that primarily regulates the tumor suppressor p53. It binds to p53, inhibiting its transcriptional activity and promoting its degradation, forming an autoregulatory negative feedback loop. MDM2 also targets other proteins for ubiquitination. Overexpression or amplification of MDM2 is observed in various cancers, leading to inactivation of p53-mediated apoptotic and cell cycle arrest functions.
