mTORC1 is a multi-protein complex that acts as a central regulator of cell growth, metabolism, and survival. It functions as a nutrient, energy, and redox sensor to control protein synthesis and other anabolic processes in response to various intracellular and extracellular signals. It is composed of mTOR, Raptor, mLST8, PRAS40, and DEPTOR. It is activated by nutrients, growth factors, and energy sufficiency. mTORC1 promotes translation by phosphorylating downstream effectors such as 4E-BP1 and S6K1. It is inhibited by rapamycin, which binds FKBP12 and inhibits mTORC1 kinase activity.
