Overview

The medial knee compartment is an anatomical region of the knee joint comprising the medial femoral condyle, the medial tibial plateau, and the medial meniscus (StatPearls, 2023). It is the most common site for the development of osteoarthritis because it bears approximately 60-70% of the total load across the knee during normal gait and weight-bearing activities (PubMed, 2021). While it is not a molecular target such as a receptor or enzyme, it is a primary focus for localized therapeutic interventions, including intra-articular injections of corticosteroids, viscosupplements, and emerging disease-modifying osteoarthritis drugs (DMOADs) (NIH, 2022). Pathological changes in this compartment, such as cartilage thinning, osteophyte formation, and meniscal damage, serve as key clinical indicators for disease progression and are used as primary endpoints in orthopedic clinical trials (Arthritis & Rheumatology, 2020). Consequently, the medial knee compartment is a critical site of action for drugs intended to alleviate joint pain and preserve structural integrity in degenerative joint diseases.
