Overview

Mediator complex subunit 10 (MED10) is a protein component of the Mediator complex, a large multisubunit complex that acts as a transcriptional coactivator required for the regulated transcription of nearly all RNA polymerase II-dependent genes[2][3]. MED10 is localized to the middle module of the Mediator complex and plays a crucial structural and functional role in bridging gene-specific regulatory factors and the basal transcription machinery, including RNA polymerase II. Functional studies, primarily in model organisms such as yeast and supported by human gene data, show MED10 is involved in the assembly and stability of the preinitiation complex (PIC). Mutations in MED10 (or its homologs in other species) impair gene transcription globally by affecting the interaction and recruitment of general transcription factors (notably TFIIB and RNA polymerase II)[1]. There is no evidence that MED10 itself functions as a direct therapeutic target or biomarker; it is instead a core participant in the fundamental process of gene transcription regulation[2][3][1].
