Overview

Mediator complex subunit 11 (MED11) is a protein encoded by the MED11 gene in humans and is a component of the multiprotein Mediator complex[1][5]. This complex functions as a transcriptional coactivator, bridging specific regulatory DNA-binding proteins with the RNA polymerase II transcription machinery, thereby facilitating regulated gene expression for nearly all RNA polymerase II-dependent genes[1][5]. The Mediator complex is modular, and MED11 is part of the Head module, which directly interacts with RNA polymerase II and general initiation factors[3], supporting basal transcription. MED11 is evolutionarily conserved across eukaryotes, and its structure is highly similar between yeast and mammals[2]. Mutations or disruptions in MED11 have been associated with severe neurological, developmental, and epileptic disorders in humans[1]. There are no known interacting drugs targeting MED11, and it is not currently considered a therapeutic target in pharmacology.
