Overview

Mediator complex subunit 13 (MED13) is a large protein (about 239 kDa) encoded by the MED13 gene in humans. It is a key component of the Mediator complex, a multi-protein transcriptional coactivator essential for the regulated expression of most RNA polymerase II-dependent genes. MED13 is specifically part of the cyclin-dependent kinase 8 (CDK8) kinase module of the Mediator complex, interacting with CDK8, cyclin C (CCNC), and MED12 to modulate Mediator function. The Mediator complex functions as a molecular bridge, conveying regulatory signals from DNA-bound transcription factors and nuclear receptors to the core transcriptional machinery, thereby controlling gene expression in eukaryotes. Variants in MED13 are associated with neurodevelopmental disorders, including autism spectrum disorder, intellectual disability, ADHD, and congenital heart defects, reflecting its critical regulatory role in development and transcriptional control. No direct small molecule or biologic drugs are currently known to target MED13, but its central role in gene regulation makes it a research target for understanding dysregulated transcription in development and disease.
