Overview

Mediator complex subunit 16 (MED16) is a protein located in the nucleus, where it forms part of the multi-subunit Mediator complex. This complex is a crucial transcriptional coactivator, connecting sequence-specific transcription factors to the RNA polymerase II machinery, thereby regulating the initiation of transcription for most protein-coding genes in eukaryotes. MED16 does not possess catalytic activity but contributes to the structural and functional integrity of the Mediator complex, affecting the expression of specific gene regulons in processes such as development, stress responses, and hormone signaling. In humans, mutations in MED16 are associated with certain intellectual disability syndromes.
