Overview

Mediator complex subunit 19 (MED19) is an evolutionarily conserved nuclear protein that is a core component of the Mediator complex, which bridges DNA-binding transcription factors with the RNA polymerase II transcriptional machinery[1][5]. MED19 plays a pivotal role in transcriptional activation and repression by acting as a coactivator, directly interacting with transcription factors such as Hox proteins and helping to integrate regulatory signals into the transcription of target genes[5]. MED19 is located at chromosome 11q12.1 and encodes two major isoforms: a canonical (full-length) and an alternative (shorter) variant, both of which are present in various tissues and show differential expression patterns in prostate cancer[7]. MED19 overexpression is linked to the progression, metastasis, and poor prognosis of multiple cancer types, and it has been implicated in the regulation of key signaling pathways such as AKT/mTOR, EGFR/MEK/ERK, and WNT/β-catenin[3]. Knockdown of MED19 reduces cancer cell proliferation, migration, invasion, and promotes apoptosis, suggesting a pro-tumorigenic role[3]. MED19 is thus considered a potential therapeutic target and biomarker in oncology, although the molecular details of its involvement in different cancers remain an active area of research[1][3].
