Mediator complex subunit 26 (MED26) is a protein coding gene encoding an enzyme that functions as a subunit of the Mediator complex, a multi-protein coactivator essential for transcriptional regulation by RNA polymerase II[1][7]. MED26 sets itself apart from other Mediator subunits by playing a key role in the transition from transcription initiation to elongation, acting through its N-terminal domain, which recruits elongation factors such as ELL/EAF and P-TEFb, and interacts with TFIID and pausing-related factors (including PAF, NELF, and DSIF)[1][3][4][5][6]. MED26 is also implicated in the regulation of transcription for a subset of small nuclear RNA (snRNA) genes by coordinating the recruitment of the little elongation complex (LEC)[2][6]. Functionally, it serves as a molecular switch—facilitating the release of paused RNA polymerase II into productive elongation by direct interactions with various transcriptional regulators[4][5]. MED26 is essential in multiple developmental processes such as erythropoiesis[5], though there are no widely recognized drug interactions or direct therapeutic targeting reported. Diseases linked to mutation or dysregulation of MED26 include Opitz-Kaveggia syndrome[7].
