Mediator complex subunit 28 (MED28) is a protein component of the multi-subunit Mediator complex that serves as a crucial coactivator for RNA polymerase II transcription in eukaryotic cells. MED28 acts as a bridge transmitting signals from gene-specific regulatory factors to the basal transcription machinery, assisting in the assembly of the preinitiation complex. In addition to its nuclear role, MED28 interacts cytosolically with merlin (NF2), participating in actin cytoskeleton signaling and cellular migration. MED28 is essential for peri-implantation development, maintenance and acquisition of embryonic stem cell pluripotency, and plays a role in regulating cell differentiation, especially repressing smooth muscle differentiation. Mutations or dysregulation have been associated with certain cancers (especially breast cancer, via MEK1-dependent migration), and genetic diseases such as severe congenital neutropenia and schwannomatosis. There are currently no drugs or established clinical biomarkers directly targeting MED28.
