Overview

Mediator of RNA polymerase II transcription subunit 22 (MED22) is a core component of the Mediator complex, a large multiprotein coactivator essential for the regulated transcription of most RNA polymerase II-dependent genes[1][4][6]. MED22 is structurally part of the 'head' module of the Mediator and forms a heterodimer with MED11, creating a conserved four-helix bundle that is critical for the assembly and stability of the pre-initiation complex (PIC) on gene promoters, including recruitment of basic transcriptional machinery and interaction with TFIIH[2]. MED22 plays a specialized and essential role in podocyte homeostasis; its loss leads to podocyte cellular degeneration, vacuole formation, and progressive renal failure in mouse models[1]. While MED22 has not been demonstrated as a direct drug target, it is implicated in kidney disorders such as glomerulopathies and may be a candidate for further disease research. It is not an enzyme, receptor, transporter, or ion channel, but a transcriptional regulator as part of the Mediator complex[1][2][3][4][6].
