Overview

Mediator of RNA polymerase II transcription subunit 25 (MED25) is a subunit of the multiprotein Mediator complex, which acts as a crucial coactivator bridging DNA-bound transcription factors and the basal RNA polymerase II transcription machinery[6][7]. MED25 possesses several functional domains, including the ACID (activator interaction domain), which binds various transcriptional activators and viral proteins, and a von Willebrand Factor Type A domain for interaction within the Mediator complex[3][7]. It is a key integrative hub for the regulation of transcriptional responses to endogenous and environmental signals, notably in plant hormone and stress signaling in Arabidopsis (jasmonate, abscisic acid, ethylene, and auxin pathways)[1][2][5][8]. In humans, MED25 is involved in gene regulatory networks, is implicated in cancer, and can be targeted by some viral proteins (such as from human respiratory syncytial virus)[4]. There are currently no approved drugs specifically targeting MED25, nor is it established as a clinical biomarker or direct therapeutic target, but it is of high biological interest due to its central regulatory role.
