Overview

Mediator of RNA polymerase II transcription subunit 31 (MED31) is a highly conserved subunit of the Mediator complex, a multiprotein complex that functions as a central scaffold and transcriptional coactivator in the regulated transcription of nearly all RNA polymerase II-dependent genes[3][4][5]. MED31 is essential in bridging gene-specific regulatory proteins to the basal transcription machinery, facilitating the assembly of a functional preinitiation complex with RNA polymerase II and general transcription factors[3][4]. MED31’s functions include regulation of transcription, limb development, and negative regulation of fibroblast proliferation[3][5]. It is encoded by the MED31 gene in humans and is homologous to yeast SOH1[5]. MED31 is not currently classified as a direct therapeutic target, receptor, enzyme, or transporter, and there are no known interacting drugs or mechanisms of action established for clinical therapeutics[3][5].
