Overview

Megakaryocyte-associated tyrosine-protein kinase (MATK) is a non-receptor tyrosine kinase enzyme belonging to the Csk family, encoded by the MATK gene in humans. This kinase is structurally related to Csk tyrosine kinases and contains SH2 and SH3 domains, a unique N-terminus, and lacks myristylation and certain negative regulatory phosphorylation sites. MATK plays a regulatory role in hematopoietic cell signal transduction by phosphorylating and inactivating Src family kinases, thereby acting as a negative regulator especially in T-cell proliferation. It has been implicated in pathways relevant to immune response, hematopoiesis, and certain cancers, including breast and colorectal cancer (where it may act as a tumor suppressor). MATK interacts with proteins such as CD117 (c-Kit) and TrkA and is often the subject of high-throughput kinase inhibitor screens.
