Overview

Meiotic recombination protein REC114 (REC114) is a highly conserved protein essential for the initiation of meiotic recombination. It functions as a key component of the meiotic DNA double-strand break (DSB) machinery, interacting with MEI4 and IHO1 to control the timing, localization, and frequency of DSBs required for homologous recombination during meiosis. Structurally, REC114 contains an N-terminal pleckstrin homology (PH)-like domain, a central intrinsically disordered region, and a C-terminal region required for complex formation with MEI4[1][3][4]. REC114–MEI4 heterotrimers bind DNA cooperatively, facilitating protein–DNA condensation and the formation of nuclear foci at meiotic chromosomal axes, promoting the assembly of DSB machinery[1][3][4][2]. Loss of REC114 function disrupts meiotic DSB formation, resulting in infertility due to failed gametogenesis in mice and other organisms. There is no evidence or precedent for direct pharmacological targeting of REC114; its importance is restricted to meiotic cell biology and fundamental genetic integrity[3]. REC114 is not considered a receptor, enzyme, transporter, or conventional therapeutic target. It is instead classified as an essential meiosis-specific structural and regulatory component enabling DSB formation required for genetic recombination in sexual reproduction[1][3][4].
