Melanocortin receptor 5 (MC5R) is a G protein-coupled receptor (GPCR) that binds melanocyte-stimulating hormones (α-, β-, γ-MSH) and adrenocorticotropic hormone (ACTH). It is widely expressed in both central nervous system and peripheral tissues, including sebaceous glands, skeletal muscle, and adipose tissue. MC5R plays diverse roles including regulation of lipid production, modulation of sexual behavior, immunomodulation, and energy metabolism. Agonists targeting MC5R are being explored for their ability to modulate energy metabolism, potentially useful for treating metabolic disorders like obesity or diabetes mellitus.
Agonists bind to MC5R, activating G proteins, leading to increased cAMP production and PKA activation.
