Melanocortin receptor type 5 (MC5R) is a G protein-coupled receptor activated by melanocortin peptides derived from proopiomelanocortin (POMC). It is widely expressed in the central nervous system and peripheral tissues, including sebaceous glands, preputial glands, immune cells, skeletal muscle, and white adipose tissue. MC5R regulates lipid production, pheromone production, immunological responses, and energy metabolism. It has been implicated as a potential therapeutic target for obesity, diabetes mellitus, and disorders of exocrine gland function. MRAPs can regulate MC5R’s cell surface expression, dimerization status, pharmacology, trafficking within cells, ligand binding affinity/specificity, and signal transduction efficiency.
MC5R agonists promote cAMP/PKA, Ca²⁺, and PI3K–ERK1/2 signaling pathways.
