This target consists of a peptide epitope derived from the melanoma-associated protein melanocyte glycoprotein 100 (gp100), presented by the human leukocyte antigen A2 (HLA-A2), a major histocompatibility complex (MHC) class I molecule. The gp100 peptide–HLA-A2 complex is expressed on the surface of melanoma cells and can be specifically recognized by cytotoxic T lymphocytes, enabling targeted immune responses. Immunotherapies—including TCR-engineered T cells and peptide vaccines—exploit this antigen–MHC complex for cancer treatment, particularly in melanoma. Recognition requires both tumor expression of gp100 and patient HLA-A2 positivity, and therapies targeting this complex may trigger immune-related adverse events affecting normal melanocytes[4][7][9].
