Overview

The GP100 antigenic epitope presented on MHC class II is a specialized molecular complex formed by the processing of the melanocyte-specific protein PMEL (GP100) and its subsequent display on Major Histocompatibility Complex (MHC) class II molecules. GP100 is a transmembrane glycoprotein primarily involved in the maturation of melanosomes in melanocytes and is highly overexpressed in the majority of human melanomas. While much research has focused on MHC class I-restricted epitopes for CD8+ T cell activation, MHC class II-restricted epitopes (such as gp100:44-59) are critical for recruiting CD4+ helper T cells. These CD4+ T cells play a vital role in the induction and persistence of cytotoxic CD8+ T cell responses and can also exert direct anti-tumor effects through cytokine secretion. Therapeutic strategies targeting this complex include DNA vaccines like SCIB1 and various peptide-based vaccines designed to stimulate a robust, multi-pronged immune response against metastatic melanoma. Clinical interest in this target stems from its ability to overcome immune evasion by providing necessary T-cell help within the tumor microenvironment.
