Overview

Melanogenesis enzymes are a group of specialized catalytic proteins, primarily Tyrosinase (TYR), Tyrosinase-related protein 1 (TYRP1), and Tyrosinase-related protein 2 (also known as Dopachrome tautomerase or DCT), that orchestrate the production of melanin within melanocytes (UniProt P14679, P17642). Tyrosinase serves as the rate-limiting enzyme of the pathway, initiating the process by hydroxylating L-tyrosine to L-DOPA and subsequently oxidizing it to dopaquinone (PubMed 30835092). These enzymes are critical for determining skin, hair, and eye color and provide a vital defense mechanism against ultraviolet (UV) radiation-induced DNA damage. Dysregulation or overactivity of these enzymes leads to various pigmentary disorders, such as melasma, solar lentigines, and post-inflammatory hyperpigmentation (PubMed 25611360). Consequently, they are the primary therapeutic targets for dermatological skin-lightening agents like hydroquinone, kojic acid, and thiamidol, which act by inhibiting their catalytic activity (PubChem CID 785). In oncology, these enzymes are also significant as markers and potential targets in melanoma, where their expression is often markedly upregulated.
