Overview

Melanoma antigen family A10 (MAGE-A10) is a member of the melanoma-associated antigen gene family. These genes encode proteins that share significant sequence similarity but have variable promoters and first exons, allowing differential regulation across tissues. The MAGEA genes are clustered on chromosome Xq28. MAGE-A10 is classified as a cancer-testis or tumor-associated antigen because it is typically expressed only in germline tissues but becomes aberrantly activated in various cancers—including melanoma, lung cancer, ovarian carcinoma, and urothelial malignancies—where its presence can be associated with poor prognosis and resistance to chemotherapy[1][3][6]. While the precise biological function remains unclear, it may play roles during embryonic development or contribute to tumorigenesis and progression[4]. Due to its restricted normal tissue distribution and high prevalence across multiple tumors types, MAGE-A10 has become an attractive target for experimental immunotherapies such as adoptive T cell therapies using engineered receptors that recognize this antigen on malignant cells.
