MAGE-A3 is a cancer/testis antigen overexpressed in melanoma and other cancers. It is involved in embryogenesis, germ cell development, and apoptosis regulation. In tumors, its reactivation leads to protein expression that can be targeted by immune responses. It regulates E3 ubiquitin ligases like TRIM28, promoting cell proliferation, survival, and metastasis.
Targeted by T-cell based immunotherapies, vaccines, and adoptive T-cell transfer therapies. Regulates E3 ubiquitin ligases like TRIM28 leading to degradation of tumor suppressors TP53 and AMPK. Promotes cell proliferation, survival (by repressing apoptosis), invasion/metastasis via epithelial-mesenchymal transition
