Melanoma antigen gene family A member 3 (MAGEA3) is a cancer-testis antigen normally expressed in reproductive tissues but aberrantly re-expressed in various cancers, including melanoma and non-small cell lung cancer. It interacts with STAT1 and modulates the AMPK/mTOR pathway, promoting cell viability and tumor growth. MAGE-A3 elicits immune responses and is a target for cancer vaccines and PROTAC-based therapies.
