Melanoma-associated antigens comprise a large protein family, most notably the MAGE family, which are highly expressed in malignant tumors such as melanoma and various epithelial cancers, but are generally absent from normal adult tissues except the testis and placenta. They are classified as cancer/testis antigens and are recognized by cytotoxic T cells, making them attractive as targets for cancer immunotherapy. Multiple subfamilies exist (notably MAGE-A, MAGE-B, MAGE-C), each containing several genes with some functional redundancy and variation. MAGE proteins are under investigation as therapeutic targets and as biomarkers for cancer diagnostics and prognostics. Currently, there are no drugs directly inhibiting these antigens, but immunotherapeutic approaches, including vaccines and cellular therapies, are in development.
