Overview

Melanoma-associated antigen 1 (MAGEA1) is a member of the MAGE-A subfamily of the MAGE family of proteins. It functions as a transcriptional repressor, interacting with Ski Interacting Protein and histone deacetylases. MAGEA1 is normally expressed in testis and placenta but is aberrantly expressed in various cancers, including melanoma, lung cancer, and esophageal cancer. Its tumor-specific expression and role in immune evasion make it a target for cancer immunotherapies.

Mechanism of action

Enhances TRIM28/KAP1's ubiquitin ligase activity against p53, resulting in its degradation; binds to p53 DNA-binding domain directly which may prevent its transcriptional activity.
