Overview

The melanoma-associated antigen 1 peptide presented by HLA-A*01:01 is a tumor-associated antigenic peptide derived from the MAGE-A1 protein, which is selectively presented on the surface of tumor cells in the context of the human leukocyte antigen HLA-A*01:01, an MHC class I molecule[6][7]. MAGE-A1 is a cancer/testis antigen typically restricted to germ cells and certain malignancies (such as melanoma, lung, breast, and other cancers), but not expressed in most somatic adult tissues[7]. This peptide–MHC complex is a specific target for T cell-based immunotherapies, as T lymphocytes (including genetically engineered CAR-T or TCR-T cells) can recognize and kill tumor cells that present this peptide[6]. The selectivity of antigen presentation, governed by both HLA type and tumor expression, makes it a validated therapeutic target and a biomarker for patient selection, but also introduces risks of immune-mediated toxicity if the antigen is expressed outside tumors[5][6].
