Overview

The **MAGE-A1 peptide presented by HLA-A*01:01** refers to a peptide fragment derived from the melanoma-associated antigen 1 (MAGE-A1), a cancer/testis antigen commonly expressed in a variety of tumors but not in most normal tissues. When processed intracellularly in cancer cells, this peptide binds to the HLA-A*01:01 molecule (a class I major histocompatibility complex allele) and is presented on the cell surface. This complex can be specifically recognized by cytotoxic T cell receptors engineered for cancer immunotherapy (TCR-T therapy). Such targeting holds promise for selective elimination of tumor cells in cancers expressing both MAGE-A1 and HLA-A*01:01, but therapy is contingent upon both antigen and HLA presence and may be associated with off-target toxicity due to cross-reactivity with similar peptides in normal tissues[1][2][3].
