Overview

Melanoma-associated antigen 10 (MAGE-A10) is a member of the cancer-testis antigen family, which is typically expressed only in the germ cells of the testis and is aberrantly upregulated in various malignant tumors (UniProt P43363). The specific peptide sequence spanning residues 389-397 (GLYDGREHSV) is processed intracellularly and presented on the tumor cell surface by the Human Leukocyte Antigen (HLA) alleles A*02:01 and A*02:06 (ClinicalTrials.gov NCT02858310). This peptide-MHC complex serves as a highly specific target for engineered T-cell receptor (TCR) therapies, such as ADP-A2M10 (Adaptimmune Therapeutics). Because MAGE-A10 is not expressed in normal adult tissues—and the testis lacks HLA expression—this complex provides a favorable therapeutic window for treating solid tumors like non-small cell lung cancer and urothelial carcinoma. Drugs targeting this complex utilize affinity-enhanced TCRs to recognize the complex and trigger a cytotoxic immune response, leading to the destruction of MAGE-A10-expressing cancer cells (Journal of Clinical Oncology, 2018).
