Overview

Melanoma-associated antigen 11 (MAGEA11) is a cancer/testis antigen encoded by the MAGEA11 gene, primarily located on the Xq28 chromosome. It functions as a substrate adaptor for E3 ubiquitin ligases, guiding the ubiquitination and degradation of specific target proteins, such as the tumor suppressor PCF11[1]. MAGEA11 is known to act as a transcriptional co-regulator for the androgen and progesterone receptors, modulating their signaling in hormone-responsive tissues[3]. Normally restricted to testis (germ cells), aberrant expression occurs in various cancers, most notably melanoma and prostate cancer, implicating its role in tumorigenesis and its potential as a therapeutic target or biomarker. Preclinical studies have identified small molecule inhibitors targeting the MAGEA11-substrate binding interface, indicating the feasibility of therapeutic intervention[1].
