Overview

Melanoma-associated antigen 3 (MAGE-A3) peptide–Major Histocompatibility Complex (MHC) complexes are cell-surface targets formed when the MAGE-A3 protein is processed into short peptides and presented by HLA molecules, most commonly HLA-A*01 or HLA-A*02 (UniProt, P43357). MAGE-A3 is a prominent member of the cancer-testis antigen family, characterized by high expression in various tumors—such as melanoma, non-small cell lung cancer, and head and neck squamous cell carcinoma—while being absent in normal tissues except for the immune-privileged testes (Vansteenkiste et al., 2013). This tumor-specific presentation makes the MAGE-A3–MHC complex a primary target for T-cell receptor (TCR) engineered T-cell therapies and therapeutic cancer vaccines. The therapeutic goal is to enable T cells to recognize these specific complexes, leading to the targeted destruction of malignant cells. However, clinical development has been hindered by severe safety concerns, including lethal cardiotoxicity caused by TCR cross-reactivity with the muscle protein Titin and neurotoxicity due to cross-reactivity with MAGE-A12 in the central nervous system (Linette et al., 2013; Morgan et al., 2013). Despite these challenges, the complex remains a significant focus for refined immunotherapy approaches that prioritize high specificity to avoid off-target effects.
