Overview

Melanoma-associated antigen 6 (MAGEA6) is a member of the MAGE (Melanoma Antigen Gene) family of cancer-testis antigens, typically only expressed in adult germ cells but aberrantly activated in a wide range of cancers including melanoma, lung, colon, breast, and multiple myeloma[1][3][4][5]. MAGEA6 encodes a protein of 314 amino acids that plays a role in promoting tumorigenesis by modulating key pathways such as downregulating the tumor suppressor AMPK (through interaction with an E3 ligase TRIM28), which leads to increased mTOR signaling, decreased autophagy, and enhanced tumor cell survival and proliferation[4]. MAGEA6 is immunogenic and is the subject of immunotherapy approaches, including peptide vaccines, TCR-based cellular therapies, and may act as a biomarker for the effectiveness of AMPK agonists (like metformin) and mTOR inhibitors (like everolimus) in cancer treatment[3][4]. Safety concerns center on the potential for immune targeting of non-tumor tissues if expression occurs outside the immune privileged germline compartment. There are no small-molecule antagonists in clinical use directly targeting MAGEA6, but immune therapies targeting the antigen or related family members are in clinical and preclinical development[3][4].
