Overview

The Melanoma-associated antigen 8 (MAGEA8) peptide–HLA class I complex is a tumor-specific antigen assembly presented on the surface of various malignant cells (UniProt: P43364). MAGEA8 belongs to the cancer-testis antigen (CTA) family, which is characterized by restricted expression in the immune-privileged environment of the testis and aberrant overexpression in multiple cancers, including melanoma and lung carcinoma (PubMed: 11007144). The complex consists of a processed MAGEA8-derived peptide, such as the HLA-A*02:01-restricted epitope GLYDGREHSV, bound within the groove of a Human Leukocyte Antigen (HLA) class I molecule (PubMed: 11807001). This presentation allows the immune system, specifically CD8+ T cells, to identify and eliminate cancerous cells. Therapeutic strategies targeting this complex include the development of T-cell receptor (TCR) engineered T-cell therapies and TCR-like antibodies designed to bypass central tolerance and induce a potent anti-tumor response. Because MAGEA8 is not expressed in normal somatic tissues, it serves as a highly specific target for precision immunotherapy.

Mechanism of action

T-cell receptor (TCR) mediated recognition of the peptide-HLA complex on the tumor cell surface, leading to the formation of an immunological synapse, secretion of cytotoxic granules such as perforin and granzymes, and induction of apoptosis in the target cancer cell.
