Melanoma-associated antigen 6 (MAGEA6) is a cancer/testis antigen belonging to the MAGEA gene family. It is normally expressed in germ cells of the testis, but is aberrantly re-expressed in various cancers, including melanoma, head and neck squamous cell carcinoma, lung carcinoma, breast carcinoma and gastric cancer. MAGEA6 functions as a ubiquitin ligase that targets AMP-activated protein kinase (AMPK) for degradation, impacting cellular metabolism regulation and potentially promoting tumorigenesis. Due to its restricted expression profile, MAGEA6 is considered an attractive target for cancer immunotherapy.
MAGEA6 acts as an activator of ubiquitin ligase activity, specifically targeting AMPK for degradation, thereby suppressing AMPK-mediated apoptosis and promoting tumorigenesis.
