Overview

Melanoma-associated antigen A10 (MAGE-A10) is a cancer/testis antigen primarily localized in the nucleus. It is normally expressed in immune-privileged sites such as the testis and placenta, but becomes aberrantly upregulated in various human solid tumors, making it an attractive target for immunotherapy. Clinically, MAGE-A10 has been targeted by cancer vaccines and adoptive cell therapies, such as ADP-A2M10, demonstrating some disease stabilization in clinical trials. However, challenges remain due to overlapping antigen expression with related proteins and the need for multi-antigen strategies.

Mechanism of action

Engineered T-cells recognize HLA-A*02:01-restricted peptides from MAGE-A10, leading to targeted cytotoxicity of MAGE-A10 expressing cells.
