Overview

Melanoma-associated antigen A3 (MAGE-A3) is a tumor-specific protein encoded by the MAGEA3 gene. It is a cancer/testis antigen with restricted expression in normal tissues (testes and placenta) but is frequently overexpressed in various malignancies. MAGE-A3 acts as a cellular master regulator by interacting with E3 ubiquitin ligase TRIM28/KAP1, influencing cellular targets such as p53 and AMPK. It is considered an attractive target for immunotherapy due to its tumor specificity and is also a prognostic marker associated with poorer outcomes in several cancers.
