Overview

Melanoma-associated antigen B1 (MAGEB1) is a member of the MAGE-B family of cancer-testis antigens, a group of proteins typically expressed in the testis but aberrantly activated in a variety of cancers, including melanoma and carcinomas of the colon, esophagus, and stomach[1][3]. MAGEB1 is encoded by a gene located on chromosome Xp21 and shows substantial sequence homology with other MAGE family members[3]. It is not expressed in most normal tissues other than testis but is frequently upregulated in malignancies, likely through epigenetic mechanisms such as DNA hypomethylation[1]. Its expression is associated with tumor cell survival, regulation of apoptosis, and immune system recognition, making it a candidate biomarker for cancer diagnosis and prognosis, and a potential target for immunotherapeutic approaches[1][2][3]. Currently, there are no approved drugs that directly target MAGEB1, but similar cancer-testis antigens are the focus of vaccine and T-cell therapy development[2].
