Melanoma-associated antigen B18 (MAGEB18) is a cancer-testis antigen and member of the type I MAGE protein family, encoded on the X chromosome and primarily expressed in testis under normal conditions, but aberrantly upregulated in various tumors including melanoma. MAGEB18 is proposed to play a regulatory role in ubiquitination by binding to RING-type E3 ubiquitin ligases (such as LNX1), potentially modulating their substrate specificity or ligase activity and thus impacting protein stability, signaling pathways, and tumorigenesis. As with other members of the MAGE family, altered expression of MAGEB18 may be associated with poor clinical prognosis in cancers and is being studied as a potential immunotherapy target and biomarker.
