Overview

Melanoma-associated antigen B2 (MAGEB2) is a member of the type I cancer-testis antigen family, encoded by the MAGEB2 gene on the X chromosome (Xp21.2). It is characterized by a conserved MAGE homology domain and is normally expressed in testis and placenta but aberrantly expressed in a wide variety of malignancies. Functionally, MAGEB2 promotes tumor cell proliferation via a p53-independent pathway, mainly by enhancing E2F transcription factor activity through inhibition of HDAC1-mediated repression. MAGEB2 also localizes to the nucleolus and contributes to ribosomal biogenesis by enhancing rRNA transcription, which supports the high protein synthesis demands of proliferating cancer cells. Its expression is linked to increased tumor growth, invasion, and poor prognosis, making it a promising immunotherapy target and biomarker in multiple cancers. No small-molecule or approved drugs currently target MAGEB2 directly; its therapeutic potential primarily lies in antigen-directed immunotherapies.

Mechanism of action

Current therapeutic investigation focuses on use as an immunotherapy antigen (vaccine target). Drugs, where developed, would likely act by inhibiting its protein-protein interactions or immune-based (T cell) targeting.
