Melanoma-associated antigen B3 (MAGEB3) is a member of the cancer/testis antigen family, typically expressed in the germ cells of testis but aberrantly activated in various malignancies, notably melanoma, lung, breast, colon, and multiple myeloma. MAGEB3, like other Type I MAGE antigens, binds to and modulates specific E3 RING ubiquitin ligases through its conserved MAGE homology domain, contributing to the polyubiquitination and proteasomal degradation of tumor suppressors, including p53. Through this function, MAGEB3 promotes oncogenic processes such as increased cell proliferation, inhibition of apoptosis, and potentially drives resistance to stress and therapy in cancer cells.
