Overview

Melanoma-associated antigen B3 (MAGEB3) is a member of the cancer/testis antigen family, typically expressed in the germ cells of testis but aberrantly activated in various malignancies, notably melanoma, lung, breast, colon, and multiple myeloma. MAGEB3, like other Type I MAGE antigens, binds to and modulates specific E3 RING ubiquitin ligases through its conserved MAGE homology domain, contributing to the polyubiquitination and proteasomal degradation of tumor suppressors, including p53. Through this function, MAGEB3 promotes oncogenic processes such as increased cell proliferation, inhibition of apoptosis, and potentially drives resistance to stress and therapy in cancer cells. Its tumor-restricted expression and immunogenicity make it a relevant therapeutic and diagnostic target, particularly for cancer immunotherapy approaches, though immunological safety concerns exist due to the potential for testicular antigen cross-reactivity[1][3][5].
