Overview

Melanoma-associated antigen C2 (MAGE-C2), also known as CT10, is a member of the cancer-testis antigen (CTA) family, which is characterized by restricted expression in the testis and high expression in various malignancies such as melanoma, prostate cancer, and multiple myeloma (UniProt Q9UBF1). The MAGE-C2 peptide presented on Human Leukocyte Antigen B*07:02 (HLA-B*07:02) is a specific peptide-MHC (pMHC) complex, typically involving the decapeptide SPRWWPTCL, that serves as a target for immunotherapy (PubMed: 11160695). This complex is formed when intracellular MAGE-C2 proteins are processed by the proteasome and the resulting peptides are loaded onto HLA-B*07:02 molecules for presentation on the cell surface. Because MAGE-C2 is not expressed in essential healthy tissues, this pMHC complex is an ideal target for T-cell receptor (TCR) engineered T-cell therapies and TCR-like bispecific antibodies. These therapeutic approaches aim to redirect the patient's immune system to recognize and lyse tumor cells presenting the MAGE-C2/HLA-B*07:02 complex. Clinical interest in this target is driven by the need for highly specific cancer antigens that minimize off-tumor toxicity while providing a broad therapeutic window in HLA-B*07:02 positive patients.
