Overview

Melanoma-associated antigen D1 (MAGED1) is a member of the MAGE family, distinct from more cancer-restricted MAGE-A/B/C subfamilies. Unlike those, MAGED1 is expressed in almost all normal adult tissues and participates in multiple biological processes such as apoptosis (via p75 neurotrophin receptor), cell cycle regulation, circadian rhythm, and modulation of transcription factor activity and gene expression through epigenetic mechanisms. MAGED1 is implicated in various cancers (via roles in proliferation and apoptotic signaling), neurological dysfunction (including intellectual disability and addictive behavior), and Prader-Willi syndrome. Interactions with proteins such as UNC5A, XIAP, PJA1, USP7, and members of the DLX family mediate its diverse functions. While this molecule is a biological target and modulator rather than a common drug target, its broad tissue expression and complex regulatory roles represent significant challenges for therapeutic targeting.
